How do I enter a Boss Tower?

Just started playing and have managed not to die yet but that may come to an end if I can figure out how to enter the boss tower..

Of course I pressed 'E' already but that does nothing..

The entrances of boss towers are the same is mission entrances. So, if you've entered any mission before, you are looking for the same thing. Mission entrances are always in the first chunk of the region from the map.
